Intended Use: Summary and Explanation
PureSpermŽ 80/40 is a density-gradient product designed for the separation and
purification of normal spermatazoa from human
ejaculate by centrifugation, a standard procedure in Assisted Reproduction Technology (ART).
PureSpermŽ 80 is ready to use in conjunction with PureSpermŽ 40, to provide layers in density gradients.
PureSpermŽ 80/40 has the following specifications:
- Colloidal silica particles firmly coated with silane, in an isotonic salt solution
- Stable colloid/salt solution with shelf life of 2 years from production in unopened bottle
- pH of 7.4 - 7.8 at room temperature and at +37ºC
- Isotonicity of 300 - 310 mOsm/kg water
- HEPES buffer included
- Glucose included
- Low endotoxin levels (<1.00 EU/mL)
- Terminally sterilised by autoclaving
- Carbon content in dry residue 4 - 5%
- Density of approximately 1.10 g/mL
- Packaged in borosilicate glass bottles with silicone stoppers and tamper-evident seals
- Storage of unopened bottles at ambient temperature, after opening at 4 - 8ºC

Storage and Stability
Store unopened bottles at 240ºC and avoid temperatures above
or below these values. Under these conditions PureSpermŽ 40 and
PureSpermŽ 80 have a shelf-life of 2-years. The expiry date is shown on both bottles and cartons.
Open and close bottles under aseptic conditions. After opening, store at 2-8ºC when not in use.
No antibiotics, unstable additives or preservatives have been included in PureSpermŽ 40 or PureSpermŽ 80.
Precautions and Warnings
- When retrieving the sperm pellets, follow the instructions provided overleaf to avoid inadvertent contamination.
- Use aseptic procedures at all times.
- Use sealed buckets during centrifugation to avoid creation of aerosols.
- Clean accidental spills using a dampened cloth or paper.
- PureSpermŽ 40 and PureSpermŽ 80 cause floors and benches to be extremely slippery.
- PureSpermŽ 40 and PureSpermŽ 80 do not represent any fire or combustion hazard. A material safety data sheet is available from the manufacturer or distributor.
- Federal Law (USA) restricts this device to sale by or on the order of a physician.
- It is not intended that these gradients should be used more than once, because of sperm sample mixing and reduced gradient efficacy.
